T1 Effects of Dietary ω-3 Polyunsaturated Fatty Acids on Myocardial Mitochondria Functioning under Isopreterenol-induced Heart Damage JF International Journal of Physiology and Pathophysiology JO IJPP YR 2015 FD 2015-03-16 VO 6 IS 1 SP 69 OP 77 K1 mitochondria K1 ω-3 polyunsaturated fatty acids K1 isopreterenol AB We studied the effect of ω-3 polyunsaturated fatty acids (ω-3 PUFA, a drug Epadol, 0.1mg/100 g body weight for 4 weeks) on the functioning of the rat myocardium mitochondria under isopreterenol-induced injury (two subcutaneous injections of isopreterenol at the dose 60 mg/kg/day). It has been shown that isoproterenol-induced damage to the heart leads to respiratory failure in isolated mitochondria in the presence of succinate. An administration of ω-3 PUFAs significantly restores the respiration rate of the mitochondria: ADP-stimulated (V3) respiration rate increases by 70.1%, the rate of phosphorylation in a relatively steady state (V2) increases by 39.9%, and the respiratory control ratio − by 45.19% compared to rats injected subcutaneously with isoproterenol solution. In addition, the ability of ω-3 PUFAs to decrease mitochondrial swelling (by 60%) in nominally calcium free solution has been shown. The results obtained indicate that ω-3 PUFAs reduce mitochondrial dysfunction in the heart under isopreterenol-induced damage to the latter.
