THERMAL STUDY OF LAB ON CHIP (LOC) SYSTEM FOR PCR REACTION APPLICATIONS

RESUMO

In this paper, the thermal study of Polydimethylsiloxane (PDMS) based lab on chip (LoC) system on a glass substrate for polymerase chain reaction (PCR) has been presented. The system consist of a microfluidic structure on one side of 5×5 cm2 of glass substrate comprises of a microchannel channel with inlet and outlet holes for the fluid handling, a reaction chamber for the thermal cycle of polymerase chain reaction (PCR), and two thermally actuated membrane-type microvalves to confine the water based biological solution. While on the other side of glass substrate three thin film Cr/Al/Cr metal heaters have been integrated. Two lateral heaters, located under the valves near the inlet and outlet positions of the chamber, actuate the valves and allow the chamber isolation, while the last one, positioned below the chamber, is dedicated to the PCR thermal cycle. In order to optimize heaters and valves geometry, the temperature distribution over the heaters and the membrane of valve deformation have been studied using the commercial software COMSOL Multiphysics. The heaters have been fabricated on glass substrate according to simulated structures using soft lithography. In order to check the reliability of structure, thermal study of fabricated heaters have been performed using FLIR IR camera, which shows a good agreement with simulated values.
