Isolation and purification of sulfated anticoagulant compound from fermented Irish moss (Chondrus ocellatus)

Natural fermentation was carried out to extract the soluble polysaccharide containing anticoagulant activity from red seaweed Chondrus ocellatus. Polysaccharide having anticoagulant activity was purified by ion exchange followed by gel filtration chromatography. The anticoagulant activity was determined by activated partial thromboplastin time (APTT), prothrombin time (PT), and thrombin time (TT) assays. The polyacrylamide gel electrophoresis showed that the molecular mass of purified polysaccharide was 500 kDa and a single spot on agarose gel confirmed that the compound was purified. The concentration of the purified anticoagulant was 360 μg/mL and it showed more than 1000 (>1000) APTT and TT activity at the concentration of 162.0 μg/mL. However, the purified polysaccharide showed weaker anticoagulant activity compared to the heparin (183 IU/mg) equivalents. The purified anticoagulant showed 17% protein and 29.6% sulfate content suggesting that the compound is sulfated proteoglycan. As polysaccharides from seaweeds are good alternative sources for anticoagulant activity and can be used in a technique of fermentation because it is inexpensive and available, this purified anticoagulant polysaccharide compound could be used in pharmaceutical and biomedical industry.
