Mushrooms are a popular food with well-known functional properties, including immunoregulatory, antitumor, and antibacterial activities. In this study, we screened the anti-inflammatory and antimicrobial activities of 44 wild mushrooms growing on the campus of Rakuno Gakuen University in Japan from September to October 2013. The mushroom species were identified by morphological analysis, rDNA sequencing, and/or protein profiling. Hot-airdried powder samples were extracted with water, 50% ethanol, or hexane. Anti-inflammatory activity was evaluated by assessing the inhibition of nitric oxide (NO) production in RAW264 murine macrophages when the extracts were incubated with lipopolysaccharide. Antimicrobial activity was measured using turbidimetry based on the growth inhibition of Escherichia coli and Staphylococcus aureus cultured with the extracts. Ten mushroom samples from five species (Naematoloma fasciculare, Cortinarius balteatocumatilis, C. triumphans, Russula rosacea, and Pholiota mixta) exhibited significant anti-inflammatory effects and depressed NO production to less than 20% of that observed in the control. By contrast, only four C. balteatocumatilis strains strongly inhibited bacterial growth to less than 20% of that observed in the control. Additional C. balteatocumatilis fruiting bodies collected in 2014 and 2015 exhibited similar activities. We further focused on the identification of the responsible components and the mechanisms underlying their functional properties. Interestingly, specific ethanol extract fractions with anti-inflammatory activity were isolated, whereas many water extract fractions contained a variety of candidates with antimicrobial activity.
