Optimum Temperature and pH for Mycelial Growth of Cordyceps nutans Pat. (Ascomycetes)
ABSTRACT
Cordyceps nutans Pat. is an entomopathogenitic Ascomycete belonging to the family Clavicipitaceae and is parasitic on hemipteran insects. Very few investigations have been made with this fungus. C nutans grows very slowly. Thus, further evaluation of culture conditions was necessary. In this research, we examined optimum temperature and pH for mycelial growth of C. nutans. In order to find the optimal culture temperature for growth, three strains were incubated at 5, 10, 15, 20, 25, 30, and 35 °C in darkness for 3 months, with six repetitions. Seven initial pH conditions of the media were adjusted by using 1 N HCl or KOH, and set at every 1.0 pH level from 5.0 to 11.0. Cultures were incubated at 25 °C for 3 months in replicates of six. Mycelial growth was observed at 1025 °C and was greater at 20 and 25 °C. At 5,30, and 35 °C, mycelial growth was not observed in any of the strains. Mycelial growth was observed at pH levels from 6.0 to 11.0 and not observed at pH 5.0. The overall tendency was that the strains grew relatively well in neutral to weak alkaline pH levels (pH 7.0∼9.0) compared with acidic pH levels, but colonies were sparse.
